EPCR: EPCR plays a crucial role in maintaining normal homeostasis and regulating coagulation and inflammation.
- Gene Information: Located on mouse chromosome 2, EPCR is a multifunctional gene that has various ligands, such as Proc, and is expressed in a wide range of human cell types.
- Signaling Pathway: EPCR is a key regulator of the activated protein C anticoagulant pathway and is a type I transmembrane protein that also exists in a soluble form.
- Therapeutic Inhibition: Rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE) are associated with the EPCR pathway, making it possible to develop inhibitory antibody drugs or small nucleic acid therapeutics for targeted blockade therapy.
Targeting Strategy
B-hEPCR mice
- The exons 1-4 of mouse Procr gene which encode the signal peptide,extracellular domain were replaced by human counterparts in B-hEPCR mice.
- The promoter , 5’UTR, transmembrane and cytoplasmic domains region of the mouse gene were retained, but the 3’UTR region of the mouse gene are replaced by human counterparts.
- The human EPCR expression is driven by endogenous mouse Procr promoter, while mouse Procr gene transcription and translation will be disrupted.
EPCR mRNA expression analysis
Strain specific analysis of EPCR mRNA expression in wild-type C57BL/6JNifdc mice and B-hEPCR mice by RT-PCR. Rectum RNA was isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hEPCR mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse Epcr and human EPCR primers. Sequencing of the PCR products confirmed that the amplified sequences were consistent with database reference sequences.
EPCR Protein Expression
Western blot analysis of EPCR protein expression in homozygous B-hEPCR mice. Various tissue lysates were collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hEPCR mice (H/H), and then analyzed by Western blot with species-specific anti-EPCR antibody (Abcam: human ab300565, mouse ab302911). 40 μg total proteins were loaded for Western blotting analysis.
