• Gene Information: NRP2 (Neuropilin-2) encodes a transmembrane glycoprotein that serves as a co-receptor for VEGF and class 3 semaphorins, playing critical roles in angiogenesis, lymphangiogenesis, and axon guidance.
• Protein Expression: NRP2 is upregulated in multiple aggressive cancers (e.g., prostate, TNBC, colorectal, bladder) and correlates with poor prognosis. It is expressed on tumor cells, endothelial cells, and immune cells within the tumor microenvironment.
• Signaling Pathway: NRP2 interacts with VEGFR, c-Met, PDGFR, and integrins to activate ERK, PI3K/AKT, and p38 pathways, driving proliferation, migration, and EMT.

Strain specific analysis of NRP2 mRNA expression in wild-type C57BL/6JNifdc mice and B-hNRP2 mice by RT-PCR. Heart RNA was isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hNRP2 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human NRP2 primers.

Western blot analysis of NRP2 protein expression in homozygous B-hNRP2 mice. Various tissue lysates were collected from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-NRP2 mice (H/H), and then analyzed by western blot with species-specific anti-NRP2 antibody (abcam, ab273584). 40 μg total proteins were loaded for western blotting analysis. antibody was cross-reactive between human and mouse.

Soluble human NRP2 expression in wild-type C57BL/6JNifdc and homozygous B-hNRP2 mice was evaluated by ELISA. Plasma was collected from wild-type (n=3, 8-week-old) and homozygous B-hNRP2 (n=3, 9-week-old) mice. Protein expression level of NRP2 was analyzed by ELISA (abcam, ab315040).
