Abstract
ITGB6 and B7H3 are co-overexpressed in multiple solid tumors, presenting an opportunity to address tumor heterogeneity and resistance. Using the RenLite mouse platform, we generated a bispecific antibody (1+1 Format, Common light chain + KiH) and conjugated it with complementary Topoli and MTi payloads to create BCG048. BCG048 showed potent antitumor activity in PDX models insensitive to single-payload ADCs. The safety profile has also been preliminarily validated in cynomolgus macaques. BCG048 may overcome primary resistance and delay acquired resistance compared to traditional ADCs.
