    Title: Advancing CNS Drug Discovery: Innovative BBB Shuttle Antibodies and Humanized Models

    Despite decades of successful antibody and RNA therapies for peripheral diseases, only a very small number of such successes have been achieved in CNS diseases, mainly due to the presence of the blood brain barrier (BBB), which limits the CNS exposure of large molecule drugs. Among the various approaches to increase brain exposure of these agents, BBB shuttles have become an intensely studied area and have garnered strong interest in both industry and academia. This shuttle approach utilizes a BBB-targeting moiety to carry therapeutic cargo across the BBB via receptor-mediated transcytosis, most often in the form of bispecific antibodies, antibody fusion proteins, or antibody oligonucleotide conjugates (AOCs). Among the popular targets for BBB shuttles are transferrin receptor 1 (TfR1/CD71) and CD98hc. 

    In this webinar, we will describe our antibody discovery programs targeting these receptors, which have enabled the successful construction of BBB shuttles that demonstrate higher BBB transcytosis activity compared with benchmark molecules. RenMice®, Biocytogen’s transgenic mouse platforms used to generate novel fully human antibodies for BBB shuttles, will also be introduced.

    We will also highlight advanced humanized mouse models as powerful translational tools that support the evaluation and optimization of BBB delivery strategies and accelerate the development of CNS therapeutics, featuring models targeting TfR1, CD98hc, and IGF1R.
