    Abstract

Preclinical evaluation of CD3/CD20 T-cell engagers (TCEs) requires physiologically relevant humanized models. In humanized B-hCD3EDG/hCD20 mice and huHSC B-NDG-hlL15 mice, TCE administration induced rapid, profound peripheral B-cell clearance and completely abated serum KLH-lgG/lgM titers in TDAR assays. In OVA-induced asthma, TCEs suppressed BALF eosinophils, total lgE, and Th2 cytokines (IL-4/IL-5/IL-13). These humanized mouse platforms effectively validate CD3/CD20 TCEs for autoimmune therapeutics.
