mRNA-Based Therapeutics Summit 2026: Novel Anti-CD98hc Antibodies as Blood-Brain Barrier Penetrating Vehicles

    Abstract:

The blood-brain barrier (BBB) severely restricts macromolecular drug delivery to the brain. To overcome this, we engineered CD98hc-targeting heavy-chain-only (HcAb) and common light-chain antibodies to act as BBB shuttles via receptor-mediated transcytosis. These novel anti-CD98 vectors demonstrated superior brain penetration compared with benchmark antibodies, along with excellent drug developability. Thus, they represent highly promising delivery vehicles for transporting biologic therapeutics into the central nervous system (CNS).
