    Abstract

Pancreatic adenocarcinoma (PAAD) is an aggressive cancer characterized by a dense stroma of cancer-associated fibroblasts (CAFs), which leads to rapid progression and treatment resistance. FAP is selectively expressed by CAFs in most human epithelial cancers. GPCI is aberrantly expressed and plays a significant role in PAAD and other tumors. We developed BCG026, a bispecific ADC designed to deplete both stromal and tumor cells. BCG026 demonstrated strong binding, efficient internalization, and potent efficacy in PAAD and NSCLC models, with further preclinical studies currently underway.
