Abstract
Both ITGB6 and B7H3 are promising targets for ADC therapy, as they are co-expressed in various solid tumors. The bispecific antibody (bsAb) we developed, which targets both, showed strong binding and increased internalization, suggesting effective cytotoxic delivery. Conjugating the bsAb to a dual payload formed BCG048, which exhibits potent in vivo efficacy and synergistic effects compared to single-payload ADCs. BCG048 has the potential to effectively address tumor heterogeneity and resistance, which could benefit a wider patient population. Further studies will be performed
