Targeting strategy

Gene targeting strategy for B-hCD98/Rag2 KO mice.

The exons 2-10 of mouse Cd98 gene that encode the extracellular domain were replaced by human CD98 exons 4-12 in B-hCD98/Rag2 KO mice. The exon 3 encoding the full coding sequence and 3’UTR region of mouse Rag2 gene were knocked out in B-hCD98/Rag2 KO mice, resulting in a disruption of the Rag2 gene.

Protein expression analysis in brain endothelial cells

Strain specific CD98 expression analysis in homozygous B-hCD98/Rag2 KO mice by flow cytometry.

Brain were collected from wild type (WT) mice (+/+) and homozygous B-hCD98/Rag2 KO mice (H/H;-/-), and analyzed by flow cytometry with species-specific anti-CD98 antibody after trypsin digestion. Mouse CD98 was detectable in WT mice (+/+). Human CD98 was exclusively detectable in homozygous B-hCD98/Rag2 KO mice (H/H;-/-) but not in WT mice (+/+).

Splenocytes were collected from wild type (WT) mice (+/+) and homozygous B-hCD98/Rag2 KO mice (H/H;-/-), and analyzed by flow cytometry with species-specific anti-CD98 antibody. Mouse CD98 was detectable in WT mice (+/+) and human CD98 was not detectable in WT mice (+/+). There were no T cells in homozygous B-hCD98/Rag2 KO mice.

Protein expression analysis:

Human CD98 was exclusively detectable on brain endothelial cells and NK cells of homozygous B-hCD98/Rag2 KO mice (H/H;-/-) but not in WT mice (+/+). There were no T and B cells in homozygous B-hCD98/Rag2 KO mice relative to WT mice (+/+).

Mouse CD98 was detectable on brain endothelial cells, T cells, B  cells and NK cells in WT mice (+/+).
