Gene targeting strategy for B-hHAMP mice.
The exons 1~3 of mouse Hamp gene that encodes the full-length protein was replaced by human HAMP exons 1~3 in B-hHAMP mice. -
Protein expression analysis

Strain specific HAMP expression analysis in homozygous B-hHAMP mice by ELISA. Serum were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hHAMP mice (H/H), and analyzed by ELISA with species-specific HAMP ELISA kit. Human HAMP was exclusively detectable in homozygous B-hHAMP mice but not in wild-type mice.

mRNA expression analysis

Strain specific analysis of HAMP mRNA expression in wild-type C57BL/6 mice and B-hHAMP mice by RT-PCR. Liver RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hHAMP mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human HAMP primers. Mouse Hamp mRNA were detectable in wild-type C57BL/6. Human HAMP mRNA was detectable in homozygous B-hHAMP mice.
