Gene targeting strategy for B-hIL12RB1/hIL12RB2 mice ad.
uThe extracellular region sequences of mouse Il12rb1 were replaced by human IL12RB1 counterpart gene sequences in B-hIL12RB1/hIL12RB2 mice ad.
uChimeric CDS, composed of human IL12RB2 extracellular region plus mouse Il12rb2 transmembrane and cytoplasmic region, was inserted into the mouse Il12rb2 gene exon2 in B-hIL12RB1/hIL12RB2 mice ad.

Function analysis

IL12 induced the IFN-γ production in CD4+ T cells sorted from splenocytes. Naïve CD4+ T cells were sorted from the splenocytes in the wild-type C57BL/6 mice (WT) and heterozygous B-hIL12RB1/hIL12RB2 mice ad (HE). Cells were seeded on a 96-well plate. The plate was precoated with 0.4μg/mL anti-mCD3 mAb and 0.8μg/mL anti-mCD28 mAb at 4 ℃ for 2 hours. IL12 was added to the plate. After 48 hours of culture, the production of IFN-γ in supernatants was assessed. Mouse IL12 induced the mouse IFN-γ production in wild-type C57BL/6 mice and heterozygous humanized mice. Human IL12 induced the mouse IFN-γ production only in heterozygous B-hIL12RB1/hIL12RB2 mice ad.
