Gene targeting strategy for B-hIL13RA1 mice.
The exons 2-9 of mouse Il13ra1 gene that encode the extracellular protein were replaced by human IL13RA1 exons 2-9 in B-hIL13RA1 mice. -
mRNA expression analysis

Strain specific analysis of IL13RA1 gene expression in wild-type C57BL/6 mice and B-hIL13RA1 mice by RT-PCR. Mouse IL13RA1 mRNA was detectable only in splenocytes of wild-type C57BL/6 mice (+/+). Human IL13RA1 mRNA was detectable only in homozygous B-hIL13RA1 mice (H/H), but not in wild-type mice.

Functional analysis

Assessment of IL-13RA1-mediated responses in a model of IL13 induced airway inflammation. Two doses (10 μg per mouse) of mIL13 were administered intratracheally every other day for 4 days to wild-type C57BL/6 mice (+/+) and B-hIL13RA1 mice (H/H). 24 hours after the final IL13 challenge, the mice were assessed for BALF chemokine. mIL13 strongly induced CCL11 and CCL24 expression in wild-type mice and homozygous B-hIL13RA1 mice.
