Gene targeting strategy for B-hIL17RA/hIL17RC mice. The exons 2~9 of mouse Il17ra gene that encodes the full-length protein was replaced by human IL17RA exons 2~9 in B-hIL17RA/hIL17RC mice. The chimeric IL17RC CDS was inserted after the 5’UTR of mouse Il17rc gene in B-hIL17RA/hIL17RC mice.

Strain specific IL17RA expression analysis in C57BL/6 wild-type and homozygous B-hIL17RA/hIL17RC mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 (+/+) and homozygous B-hIL17RA/hIL17RC mice (H/H, H/H), and analyzed by flow cytometry with species-specific anti-IL17RA antibody. mIL17RA was only detectable in wild-type mice and hIL17RA were only detectable in macrophages of spleen from homozygous mice.

Bone marrow cells were collected from wild-type C57BL/6 (+/+) and homozygous B-hIL17RA/hIL17RC mice (H/H, H/H), and analyzed by flow cytometry with species-specific anti-IL17RA antibody. mIL17RA was only detectable in wild-type mice and hIL17RA were only detectable in macrophages of bone marrow from homozygous mice.

Splenocytes were collected from wild-type C57BL/6 (+/+) and homozygous B-hIL17RA/hIL17RC mice (H/H, H/H), and analyzed by flow cytometry with species-specific anti-IL17RC antibody. mIL17RC and hIL17RC were detectable in DCs, monocytes, macrophages and neutrophils from wild-type and homozygous mice, as anti-mouse IL17RC and anti-human IL17RC antibody were both cross-reactive between human and mouse.

mRNA expression analysis of IL17RC

Strain specific analysis of IL17RC mRNA expression in wild-type C57BL/6 mice and B-hIL17RA/IL17RC mice by RT-PCR. Colon RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hIL17RA/hIL17RC mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human IL17RC primers. Mouse Il17rc mRNA was detectable only in wild-type C57BL/6 mice. Human IL17RC mRNA was detectable only in homozygous B-hIL17RA/hIL17RC mice but not in wild-type mice.

Mouse Il17rc mRNA was detectable in colon of wild-type C57BL/6 mice. Human IL17RC mRNA was detectable in B-hIL17RA/hIL17RC mice.
Protein expression analysis:
Human IL17RA was exclusively detectable on macrophages in spleen and bone marrow of homozygous B-hIL17RA/hIL17RC mice, and mouse IL17RA was detectable only in wild-type C57BL/6 mice.
Human IL17RC was detectable on DCs, monocytes, macrophages and neutrophils in spleen and bone marrow of homozygous B-hIL17RA/hIL17RC mice, and mouse IL17RC was detectable in wild-type C57BL/6 mice.
