Gene targeting strategy for B-hFZD7 mice.
The exon of mouse Fzd7 gene that encodes the full-length protein was replaced by human FZD7 exon in B-hFZD7 mice. -
Protein expression analysis

Western blot analysis of FZD7 protein expression in homozygous B-hFZD7 mice. Various tissue lysates were collected from wild-type C57BL/6 (+/+) mice and homozygous B-hFZD7 (H/H) mice, and then analyzed by western blot with anti-FZD7 antibody. 40 μg total proteins were loaded for western blotting analysis. FZD7 was detected in heart, brain and skeletal muscle. The antibody cross recognises human and mouse FZD7.

mRNA expression analysis

Strain specific analysis of FZD7 mRNA expression in wild-type C57BL/6 mice and B-hFZD7 mice by RT-PCR. Brain RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hFZD7 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human FZD7 primers. Mouse Fzd7 mRNA was detectable only in wild-type C57BL/6 mice. Human FZD7 mRNA was detectable only in homozygous B-hFZD7 mice but not in wild-type mice.
