Targeting strategy
-
Gene targeting strategy for B-hF11R mice.
The exons 2~7 of mouse F11r gene that encode the extracellular domain were replaced by human F11R exons 2~7 in B-hF11R mice. -
Protein expression analysis of F11R(JAM1) in platelet
-
Strain specific F11R expression analysis in homozygous B-hF11R mice by flow cytometry. Platelet were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hF11R mice (H/H), and analyzed by flow cytometry with species-specific anti-F11R antibody. Mouse F11R was detectable in platelet of wild-type C57BL/6 mice. Human F11R was exclusively detectable in platelet of homozygous B-hF11R mice but not in wild-type C57BL/6 mice.
-
Protein expression analysis of F11R(JAM1) in T cells, B cells and Monocytes of splenocytes
-
Strain specific F11R expression analysis in homozygous B-hF11R mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hF11R mice (H/H), and analyzed by flow cytometry with species-specific anti-F11R antibody. Mouse F11R was not detectable in T cells (A), B cells (B) and monocytes (C) of wild-type C57BL/6 mice. Human F11R was not detectable in T cells (A), B cells (B) and was detectable monocytes (C) of homozygous B-hF11R mice.
-
Protein expression analysis of F11R(JAM1) in Endothelial cells of lung
-
Strain specific F11R expression analysis in homozygous B-hF11R mice by flow cytometry. Lung were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hF11R mice (H/H), and analyzed by flow cytometry with species-specific anti-F11R antibody. Mouse F11R was detectable in endothelial cells of wild-type C57BL/6 mice. Human F11R was exclusively detectable in endothelial cells of homozygous B-hF11R mice but not in wild-type C57BL/6 mice.

Summary
-
Protein expression analysis:
Human F11R was detectable in NK, DC, monocytes, macrophages, platelet, stem cells and endothelial cells of homozygous B-hF11R mice, and mouse F11R was detectable in NK, DC, macrophages, platelet, stem cells and endothelial cells of wild-type C57BL/6 mice.
