Targeting strategy

Gene targeting strategy for B-hITGB3 mice.
The signal peptide, extracellular region of human ITGB3 gene and the transmembrane and cytoplasmic region of mouse Itgb3 gene were constructed into a chimeric CDS vector and inserted into the mouse exon 1. The targeted mice will express the chimeric ITGB3 protein, while mouse ITGB3 will no longer express.

Protein expression analysis


Strain specific hITGB3 expression analysis in homozygous B-hITGB3 mice by flow cytometry. Blood platelet was collected from wild-type mice (+/+) and homozygous B-hITGB3 mice (H/H) and analyzed by flow cytometry with species-specific anti-ITGB3 antibody. Mouse ITGB3 was detectable in wild-type mice. Human ITGB3 was detectable in homozygous B-hITGB3 mice.

mRNA expression analysis

Strain specific analysis of ITGB3 mRNA expression in wild-type C57BL/6 mice and B-hITGB3 mice by RT-PCR. Spleen RNA was isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hITGB3 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human ITGB3 primers. Mouse ITGB3 mRNA was detectable only in wild-type C57BL/6 mice. Human ITGB3 mRNA was detectable only in homozygous B-hITGB3 mice but not in wild-type mice.

mRNA expression analysis:
Mouse Itgb3 mRNA was detectable in wild-type C57BL/6 mice. Human ITGB3 mRNA was detectable in homozygous B-hITGB3 mice.

Protein expression analysis:
Human ITGB3 was detectable on blood platelet, T cells, B cells, granulocytes, monocytes and macrophages of homozygous B-hITGB3 mice, and mouse ITGB3 was detectable only in wild-type C57BL/6 mice.
