Gene targeting strategy for B-hITGB3 mice.
The signal peptide, extracellular region of human ITGB3 gene and the transmembrane and cytoplasmic region of mouse Itgb3 gene were constructed into a chimeric CDS vector and inserted into the mouse exon 1. The targeted mice will express the chimeric ITGB3 protein, while mouse ITGB3 will no longer express. -
Protein expression analysis

Strain specific hITGB3 expression analysis in homozygous B-hITGB3 mice by flow cytometry. Blood platelet was collected from wild-type mice (+/+) and homozygous B-hITGB3 mice (H/H) and analyzed by flow cytometry with species-specific anti-ITGB3 antibody. Mouse ITGB3 was detectable in wild-type mice. Human ITGB3 was detectable in homozygous B-hITGB3 mice.

mRNA expression analysis

Strain specific analysis of ITGB3 mRNA expression in wild-type C57BL/6 mice and B-hITGB3 mice by RT-PCR. Spleen RNA was isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hITGB3 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human ITGB3 primers. Mouse ITGB3 mRNA was detectable only in wild-type C57BL/6 mice. Human ITGB3 mRNA was detectable only in homozygous B-hITGB3 mice but not in wild-type mice.
