Gene targeting strategy for B-hITGB7 mice. Exons 2-14 of the mouse Itgb7 gene (encoding the extracellular domain) were replaced by human ITGB7 exons 3-15 in B-hITGB7 mice.

mRNA Expression Analysis

Strain specific analysis of ITGB7 gene expression in wild-type mice and B-hITGB7 mice by RT-PCR. Mouse Itgb7 mRNA was detectable in splenocytes of wild-type mice (+/+). Human ITGB7 mRNA was detectable only in homozygous B-hITGB7 but not in wild-type mice.

Strain-specific ITGB7 expression analysis in homozygous B-hITGB7 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice and homozygous B-hITGB7 mice (H/H), and analyzed by flow cytometry with species-specific anti-ITGB7 antibody. Mouse LPAM-1(α4β7) was detectable in wild-type mice and homozygous B-hITGB7 mice (H/H). Human ITGB7 was exclusively detectable in homozygous B-ITGB7 mice but not in wild-type mice.
