Targeting strategy

Gene targeting strategy for B-hMRC1 mice.
The exons 1~30 of mouse Mrc1 gene that encode the extracellular domain were replaced by human MRC1 exons 1~30 in B-hMRC1 mice.

Protein expression analysis

Strain specific MRC1 expression analysis in homozygous B-hMRC1 mice by flow cytometry. Peritoneal exudative macrophages(PEMs) were collected from wild-type mice (+/+) and homozygous B-hMRC1 mice(H/H), and analyzed by flow cytometry with species-specific anti-MRC1 antibody. Mouse MRC1 was detectable in wild-type mice and homozygous B-hMRC1 mice as the immunogen of the antibody is the intracellular domain. Human MRC1 was detectable in homozygous B-hMRC1 mice.

mRNA expression analysis

Strain specific analysis of MRC1 gene expression in wild-type mice and B-hMRC1 mice by RT-PCR. Mouse Mrc1 mRNA was detectable in spleen of wild-type mice (+/+). Human MRC1 mRNA was detectable only in homozygous B-hMRC1 but not in wild-type mice.

Summary

mRNA expression analysis:
Mouse Mrc1 mRNA was detectable in spleen of wild-type C57BL/6 mice. Human MRC1 mRNA was detectable in B-hMRC1 mice.

Protein expression analysis:
