Gene targeting strategy for B-hRTN4R mice. The mouse Rtn4r gene that encode the full-length protein does not include the signal peptide was replaced by human RTN4R counterpart gene in B-hRTN4R mice.

mRNA expression analysis

Strain specific analysis of RTN4R gene expression in wild-type mice and B-hRTN4R mice by RT-PCR. Mouse Rtn4r mRNA was detectable in brain of wild-type mice (+/+). Human RTN4R mRNA was detectable only in homozygous B-hRTN4R mice (H/H) but not in wild-type mice.

Protein expression analysis

Strain specific RTN4R expression analysis in homozygous B-hRTN4R mice by western blot. Brain tissue and liver tissue were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hRTN4R mice (H/H) and analyzed by western blot with anti-hRTN4R antibody. Human RTN4R was detectable in brain of wild-type mice and homozygous B-hRTN4R mice, as the antibody is crossly reactive with RTN4R in human and mice.
