Description
The mouse Cd20 gene was replaced by human CD20 coding sequence in B-hCD20-luc GFP A20 cells. Human CD20 is highly expressed on the surface of the cell. And it expresses firefly luciferase and GFP as a marker.

Gene targeting strategy for B-hCD20-luc GFP A20 cells. The exogenous promoter, human CD20 coding sequence and luciferase-GFP was inserted to replace part of murine Cd20 gene in B-hCD20-luc GFP A20 cells. Mouse Cd20 transcript was disrupted, the protein no longer be expressed.

Protein expression analysis

CD20 expression analysis in B-hCD20-luc GFP A20 cells by flow cytometry. Single cell suspensions from wild-type A20 and B-hCD20-luc GFP A20 cultures were stained with anti-CD20 antibody. Human CD20 was detected on the surface of B-hCD20-luc GFP A20 cells but not wild-type A20 cells. And mouse CD20 was not detected on B-hCD20-luc GFP A20 cells. And it expresses firefly luciferase and GFP as a marker.

Luminescence signal intensity of B-hCD20-luc GFP A20 cells. Luminescence intensity was measured using the Bright-GloTM luciferase Assay (Promega, Cat E2610). B-hCD20-luc GFP A20 cells have strong luminescence signal.
