Gene targeting strategy for B-hANTXR2 mice.
The exons 1-11 of mouse Antxr2 gene that encode the extracellular domain were replaced by human ANTXR2 exons 1-11 in B-hANTXR2 mice. -
mRNA expression analysis

Strain specific analysis of ANTXR2 gene expression in wild-type C57BL/6 mice and B-hANTXR2 mice by RT-PCR. Mouse Antxr2 mRNA was detectable in wild-type C57BL/6 mice (+/+). Human ANTXR2 mRNA was detectable only in homozygous B-hANTXR2 mice (H/H), but not in wild-type mice.

Protein expression analysis

Strain specific ANTXR2 expression analysis in homozygous B-hANTXR2 mice by flow cytometry. Lung endothelial cells were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hANTXR2 mice (H/H), and analyzed by flow cytometry with species-specific anti-hANTXR2 antibody. Human ANTXR2 was exclusively detectable in homozygous B-hANTXR2 but not in wild-type mice.
