Gene targeting strategy for B-hCAIX mice. Exons 1-9 of the mouse Car9 gene that encode the extracellular domain were replaced by human CAIX exons 1-9 in B-hCAIX mice.

Species-specific CAIX gene expression analysis in wild-type and humanized B-hCAIX mice by RT-PCR. Mouse Car9 mRNA was detected in stomach tissue isolated from wild-type C57BL/6 (+/+) mice, while human CAIX mRNA was detected in homozygous B-hCAIX (H/H) mice.

Immunohistochemical (IHC) analysis of CAIX expression in wild-type and humanized B-hCAIX mice. Stomach and liver tissues were isolated from wild-type C57BL/6 and homozygous B-hCAIX (H/H) mice and analyzed by IHC using an anti-human CAIX antibody. Human CAIX was detected in B-hCAIX mice but absent in wild-type mice. Red arrows indicate tissue cells with positive CAIX staining.
