Basic Information
-
Targeting strategy
-
Gene targeting strategy for B-hCFP mice.
The exon 1-9 of mouse Cfp gene that encodes the full-length protein was replaced by human CFP exon 1-9 in B-hCFP mice. -
Protein expression analysis
-
Strain specific CFP expression analysis in homozygous B-hCFP mice by ELISA. Plasma was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCFP mice (H/H), and analyzed by ELISA with a species-specific human CFP ELISA kit. Human CFP was exclusively detectable in homozygous B-hCFP mice but not in wild-type C57BL/6 mice.
-
mRNA expression analysis
-
Strain specific analysis of CFP mRNA expression in wild-type C57BL/6 mice and B-hCFP mice by RT-PCR. Spleen RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hCFP mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CFP primers. Mouse Cfp mRNA was detectable only in wild-type C57BL/6 mice. Human CFP mRNA was detectable only in homozygous B-hCFP mice but not in wild-type mice.
