Exons 3~5 of the mouse Dlk1 gene, which encodes the full-length protein, was replaced by human DLK1 exons 3~5 in B-hDLK1 mice. -
mRNA expression analysis

Strain specific analysis of DLK1 mRNA expression in wild-type C57BL/6 mice and B-hDLK1 mice by RT-PCR. Lung RNA samples were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hDLK1 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human DLK1 primers. Mouse Dlk1 mRNA was detectable in wild-type C57BL/6. Human DLK1 mRNA was detectable in homozygous B-hDLK1 mice.

Protein expression analysis

Western blot analysis of DLK1 protein expression in homozygous B-hDLK1 mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hDLK1 mice (H/H), and then analyzed by western blot with anti-DLK1 antibodies. 1 μg/μL total proteins were loaded for western blotting analysis by JESS (Bio-Techne). Human DLK1 protein was detected in embryo, placenta, fetal liver and adrenal gland.
