Strain specific EPOR expression analysis in B-hEPOR mice by flow cytometry. Bone marrow cells were collected from wild-type C57BL/6 mice(+/+) and B-hEPOR mice(H/H) , and analyzed by flow cytometry with species-specific anti-human EPOR antibody. Human EPOR was detectable on proerythroblasts, basophilic erythroblasts and polychromatic erythroblasts in B-hEPOR mice only.

mRNA expression analysis

Strain specific analysis of EPOR mRNA expression in wild-type C57BL/6 mice and B-hEPOR mice by RT-PCR. Liver RNA were isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hEPOR mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human EPOR primers. Mouse Epor mRNA was detectable in wild-type C57BL/6 mice and homozygous B-hEPOR mice (H/H). Human EPOR mRNA was only detectable in homozygous B-hEPOR mice but not in wild-type mice.
