The exons 1-17 of mouse Her3 gene that encode the extracellular domain and transmembrane domain were replaced by human HER3 exons 1-17 in B-hHER3 mice.

Species-specific HER3 gene expression analysis in wild-type and humanized B-hHER3 mice by RT-PCR. Mouse Her3 mRNA was detected in liver tissue isolated from wild-type C57BL/6 (+/+) mice, while human HER3 mRNA was detected in homozygous B-hHER3 (H/H) mice.

IHC analysis of HER3 expression

Immunohistochemical (IHC) analysis of HER3 expression in wild-type and humanized B-hHER3 mice. Liver and colon tissues were isolated from wild-type C57BL/6 (+/+) and homozygous B-hHER3 (H/H) mice and analyzed by IHC using anti-HER3 antibodies. HER3 protein was detected in wild-type and homozygous B-hHER3 mice due to the cross-reactive antibody. The arrow indicates tissue cells with positive HER3 staining (brown).
