Gene targeting strategy for B-hIBSP mice.
The exons 2~7 of mouse Ibsp gene that encodes the full-length protein was replaced by human IBSP exons 2~7 in B-hIBSP mice.

Protein expression analysis

Strain specific IBSP expression analysis in homozygous B-hIBSP mice by ELISA. Serum was isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hIBSP mice (H/H) and analyzed by ELISA with IBSP ELISA kit. Mouse IBSP was detectable in wild-type mice and homozygous B-hIBSP mice as the kit has partly cross reactions with human and mouse (A). Human IBSP was exclusively detectable in homozygous B-hIBSP mice but not in wild-type mice (B).

mRNA expression analysis

Strain specific analysis of IBSP mRNA expression in wild-type C57BL/6 mice and B-hIBSP mice by RT-PCR. Bone marrow RNA was isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hIBSP mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human IBSP primers. Mouse Ibsp mRNA was detectable only in wild-type C57BL/6 mice. Human IBSP mRNA was detectable only in homozygous B-hIBSP mice but not in wild-type mice.
