Gene targeting strategy for B-hITGA9 mice.
The signal peptide, extracellular and transmembrane region of human ITGA9 gene and the cytoplasmic region of mouse Itga9 gene were constructed into a chimeric CDS vector and replaced the mouse exon 2. The targeted mice will express the chimeric ITGA9 protein, while mouse ITGA9 will no longer express. -
Protein expression analysis

Western blot analysis of ITGA9 protein expression in homozygous B-hITGA9 mice. Various tissue lysates were collected from from wild-type C57BL/6 mice (+/+) and homozygous B-hITGA9 mice (H/H) , and then analyzed by western blot with anti-ITGA9 antibody. 40 μg total proteins were loaded for western blotting analysis.

mRNA expression analysis

Strain specific analysis of ITGA9 mRNA expression in wild-type C57BL/6 mice and B-hITGA9 mice by RT-PCR. Lung RNA was isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hITGA9 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human ITGA9 primers. Mouse Itga9 mRNA was detectable only in wild-type C57BL/6 mice. Human ITGA9 mRNA was detectable only in homozygous B-hITGA9 mice but not in wild-type mice. ?w=1181&ssl=1″ alt=”B-hITGA9 mice” />
