Gene targeting strategy for B-hITGB8 mice. The mouse Itgb8 gene that encode the extracellular domain were replaced by human ITGB8 counterpart gene in B-hITGB8 mice.

mRNA expression analysis

Strain specific analysis of ITGB8 gene expression in wild-type mice and B-hITGB8 mice by RT-PCR. Mouse Itgb8 mRNA was detectable in brain of wild-type mice (+/+). Human ITGB8 mRNA was detectable only in homozygous B-hITGB8 mice (H/H) but not in wild-type mice.

Protein expression analysis

Strain specific ITGB8 expression analysis in homozygous B-hITGB8 mice by western blot. Brain tissue and uterus tissue were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hITGB8 mice (H/H), and analyzed by western blot with anti-hITGB8 antibody. Human ITGB8 was detectable in wild-type mice and homozygous B-hITGB8 mice, as the antibody is crossly reactive with ITGB8 in human and mice.

mRNA and Protein expression analysis: Human ITGB8 mRNA was detectable in B-hITGB8 mice, and we proved that the protein could be expressed normally by western blot analysis.
