Gene targeting strategy for B-hMST1R mice.
The exons 1~12 of mouse Mst1r gene that encodes the full-length protein was replaced by human MST1R exons 1~12 in B-hMST1R mice. -
Protein expression analysis

Western blot analysis of MST1R protein expression in homozygous B-hMST1R mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hMST1R mice (H/H), and then analyzed by western blot with anti-MST1R antibody. 1 μg/μL total proteins were loaded for western blotting analysis by JESS. Human MST1R was exclusively detected in spleen, stomach, colon, lung, liver, heart and bladder in homozygous B-hMST1R mice.

mRNA expression analysis

Strain specific analysis of MST1R mRNA expression in wild-type C57BL/6 mice and B-hMST1R mice by RT-PCR. Lung RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hMST1R mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human MST1R primers. Mouse Mst1r mRNA were detectable in wild-type C57BL/6. Human MST1R mRNA was detectable in homozygous B-hMST1R mice.
