Gene targeting strategy for B-hMUC17 mice. The exons 5-10 of mouse Muc3 gene that encode the SEA domain and part of the extracellular domain were replaced by human MUC17 exons 5-10 in B-hMUC17 mice.

Protein expression analysis

Strain specific MUC17 expression analysis in homozygous B-hMUC17 mice by flow cytometry. Single-cell suspension obtained from intestinal tissue homogenates of wild type (WT) mice (+/+) and homozygous B-hMUC17 mice (H/H), and analyzed by flow cytometry with anti-human MUC17 antibody. Human MUC17 was exclusively detectable in homozygous B-hMUC17 mice (H/H) but not in WT mice (+/+).

mRNA expression analysis

Strain specific analysis of MUC17 gene expression in wild type (WT) mice and B-hMUC17 mice by RT-PCR. Mouse Muc3 mRNA was detectable only in small intestines of WT mice (+/+). Human MUC17 mRNA was detectable only in homozygous B-hMUC17 mice (H/H) but not in WT mice (+/+).
