Gene targeting strategy for B-hNTRK2 mice.
Exons 2-12 of the mouse Ntrk2 gene, which encodes the extracellular and transmembrane domain, was replaced by human NTRK2 exons 4-14 in B-hNTRK2 mice.

Protein expression analysis

Western blot analysis of NTRK2 protein expression in homozygous B-hNTRK2 mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hNTRK2 mice (H/H), and then analyzed by western blot with anti-NTRK2 antibody. 40 μg total protein was loaded for simple western analysis. Human NTRK2 was detected in brain of B-hNTRK2 mice. Mouse NTRK2 was detected in brain of wild-type C57BL/6 mice.

mRNA expression analysis

Strain specific analysis of NTRK2 mRNA expression in wild-type C57BL/6 mice and B-hNTRK2 mice by RT-PCR. Brain RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hNTRK2 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human NTRK2 primers. Mouse Ntrk2 mRNA was detectable only in wild-type C57BL/6 mice. Human NTRK2 mRNA was detectable in homozygous B-hNTRK2 mice but not in wild-type mice.
