Gene targeting strategy for B-hPODXL mice.
The exons 1~8 of mouse Podxl gene that encodes extracellular domains of protein were replaced by human PODXL exons 1~8 in B-hPODXL mice.

mRNA expression analysis

Strain specific analysis of PODXL mRNA expression in wild-type C57BL/6 mice and B-hPODXL mice by RT-PCR. Kidney RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hPODXL mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human PODXL primers. Mouse Podxl mRNA was only detectable in wild-type mice. Human PODXL mRNA was exclusively detectable in homozygous B-hPODXL mice but not in wild-type mice.

Protein expression analysis

Western blot analysis of PODXL protein expression in homozygous B-hPODXL mice. Various tissues were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hPODXL mice (H/H), and then analyzed by western blot with anti-PODXL antibody. 40 μg total proteins were loaded for western blotting analysis. GAPDH were detected as internal control. Human PODXL was detectable in heart, lung, kidney and brain in homozygous mice. M: marker.
