Targeting strategy

Gene targeting strategy for B-hSLAMF7 mice.
The exons 3~5 of mouse Slamf7 gene that encodes the full-length protein was replaced by human SLAMF7 exons 2~4 in B-hSLAMF7 mice.

Protein expression analysis

Strain specific SLAMF7 expression analysis in wild-type C57BL/6 and homozygous B-hSLAMF7 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 (+/+) and homozygous B-hSLAMF7 mice (H/H) and analyzed by flow cytometry with species-specific anti-SLAMF7 antibody. Mouse SLAMF7 was detectable only in T, B and NK cells in wild-type mice, and hSLAMF7 was exclusively detectable in T, B and NK cells in homozygous mice.

Splenocytes were collected from wild-type C57BL/6 (+/+) and homozygous B-hSLAMF7 mice (H/H) and analyzed by flow cytometry with species-specific anti-SLAMF7 antibody. Mouse SLAMF7 was detectable only in monocytes and macrophages in wild-type mice, and hSLAMF7 was exclusively detectable in monocytes and macrophages in homozygous mice.
