Generation and Validation of Humanized GARP/TGFB1 Mice for Testing Novel Anti-human GARP Antibodies
SUMMARY
Protein expression analysis:
Human GARP and TGFB1 proteins were exclusively detected in homozygous B-hGARP/hTGFB1 mice, while mouse GARP and TGFB1 proteins were detected in wild-type mice.
Immune cell analysis:
Percent of T, B, NK cells, monocyte/macrophages, and dendritic cells were similar between wild-type C57BL/6 and homozygous B-hGARP/hTGFB1 mice, demonstrating that the introduction of hGARP/hTGFB1 in place of its mouse counterpart does not change the overall development, differentiation, or distribution of these immune cell types in the spleen, lymph node leukocytes, and blood.
In vivo efficacy study:
Combination of an anti-mouse PD-1 antibody with an anti-human GARP/latent-TGFβ1 antibody controlled MC38 tumor growth in B-hGARP/hTGFB1 mice.
